Cortana from the original Halo. In the sequels she became a bit annoying as she was forced into the storyline, but in the first she was great.

I agree. She was good in the first. In fact, I liked the first game the best when it comes to storyline and such. There was plenty of room for improvement in the "I get to do this level all over again with different enemies" department, but the story was still good in my opinion.

I'm gonna be crazy and say any of the Titans in God of War, just imagine the havoc you could bestow upon the world if you were Ghaia or Cronos or any of the others that shit would be sick.

GLaDOS. I know she's an antagonist, but technically she's an NPC too, and the funniest, most well-characterized one in gaming history in my opinion.
